2.3.1 Stock Solutions

APS, 10%:
0.5 g ammonium persulfate dissolved in 5 ml water, stored in 100 µl aliquots at -25C.
EDTA, 0.5M:
186 g/l ethylenediamine tetraacetic acid, disodium salt and 10 N NaOH to pH 8.0, autoclaved.
Glucose, 20%:
20 g glucose dissolved in 100ml water, sterile filtered.
3M KAc/5M HAc:
29.4 g potassium acetate and 11.5 ml acetic acid in 100 ml of water. Autoclaved.
NaAc, 3M:
408 g CH3COONa 3H2O in 1 litre H2O. Acetic acid to pH 5.2, autoclaved.
SDS 20%:
200 g SDS (Sodium dodecyl sulfate) dissolved in 1 litre water at 60C, not autoclaved!
TAE buffer, 40Õ:
193.6 g Tris-base, 15.2 g EDTA-Na22H2O, 108.9 g Na acetate 3H2O. Acetic acid to pH 7.2, H2O to make 1 litre. Stored at room temperature.
TBE buffer, 10Õ:
60.5 g Tris-base, 30.8 g boric acid and 3.72 g EDTA-Na2 2H2O, H2O to make 1 litre. Stored at room temperature. Remade if major residues appear in solution.
Lysis buffer 10Õ:
82.9 g NH4Cl, 10 g KHCO3 and 0.37 g EDTA-Na2 2H2O, H2O to make 1 litre. pH adjusted to 7.4, autoclaved. Stored at 4C.
Gel loading buffer, 5Õ:
50% glycerol, 0.5% bromophenol blue, 0.5% xylene cyanol in 5Õ TAE.
