2.4.6 Heat Denaturation of DNA

To produce single-stranded DNA for some reactions (see section 4.2.8), the double-stranded nucleic acids had to be separated by heating to 95C for 3 to 5 minutes. By rapid cooling to 0C (crashed ice), a renaturation is inhibited. Heat denaturation also removes secondary structures, disturbing a sequencing electrophoresis.
